I used lipofectamine2000 (Thermo) and viafect(Promega) to transfect C2C12 and primary muscle stem cell. It doesn't have a very high efficiency, but also works. Now I want to ask is it possible to transfect cultured tissue (muscle spindle, embryo slice, and isolated myofiber)? Any good idea? THANKS.
If these cells are adherent cells I suggest to you use the RNAimax instead of lipofectamine2000. I just published using this technique as I telling to you with NRK52E cells, a proximal renal tubular epithelial cells, I reached over 90% of efficiency in my transfection, I know that because the mRNA and protein levels were dramatically changed when I did the transfection. When I used a specific antimiR in my cells the gene expression of that miR decrease to almost zero, while the protein which is target of this miR increased proportionally.
