I'm doing a project on optimalization of protein isolation and purification. I have heard from a colleague that the yield of the protein is greater when using a cadmium column compared to a nickel column. I did some research on the internet but didn't find anything. Does anyone have experience with HPLC using a cadmium column and would like to share it with me?
Are you sure that he or she referred to cadmium, and not cobalt? Cobalt based columns (e.g. TALON) are a well known alternative for purification of histagged proteins. They generally provide higher specificity (though sometimes lower yields). We have used them with success.

So many things depend on which specific protein you're targeting. Was your colleague referring to cobalt? Is it a His-tagged target protein? Otherwise, maybe a metal-binding protein of some sort. See link below:

I wouldn't like to use cadmium because of its toxicity, but it has been investigated for immobilized metal affinity chromatography by a few people:

Although he indeed referred to a cadmium column, I'm considering alternatives (like the cobalt column) as well. Although I think that a cobalt column could work too, I'd need to figure out to which extend the yield is lower.
