it is true that ROS overproduction can deploarize mitochondria. But as far as I know this phenomenon occurs mainly when ROS production occurs in and by mitochondria. Exogenous H2O2 does not equally distribute though your cells, so you do not know if and how much H2O2 reach the mitochondria.

Moreover, H2O2 is not the only ROS supecies that might change the mitochondrial membrane potential. You also do not know, if your antioxidants reach the mitochondria to exert their effect.

Perhaps you can give me a little bit more details of your experimental settings to help you.

Before moving into mitochondrial membrane potential assays I measure the % viability in between Control-H2O2 treated cells and H2O2+antioxidants mixtures. The first group was significally moving downwards as time went by whereas the second one was almost near the control. Moreover, I meaured ROS production with DCFDA 5μΜ and results gave significant higher values in H2O2 group.

Cell do not detach but appear rather shrunk and morphologically changed.

I use Merck milipore hydrogen peroxide 30%
