Hi all, I wonder if it is possible to distinguish mouse spleen Th1, Th2 and Th17 subsets based on surface markers with Flow Cytometry WITHOUT in vitro differentiation or maybe after a 2-3d culture. Are the percentages of these subsets so low before polarization making it impossible to be detected?

I have not tested that myself, but for Th1(CXCR3, CCR5)/Th2 (CCR3, CCR4)/Th17 (CCR6) surface markers, the below links may be helpful.

The frequencies of these subsets should depend on the memory fraction sizes and thus on the degree of antigen experience the tested mice have had. The following data sheet of a BD Bioscience has helpful sample dot plots.
