I am working on poor water soluble drugs, it is also having anticancer activity few researchers reported its anticancer activity also. I was prepared nanoparticles of that drug. My big doubt is during anticancer activity studies if the solution of anticancer drug and solution of drug nanoparticles are used then both will get dissolved then we can't say that by preparing nanoparticles its anticancer activity was enhanced. The drug and drug nanoparticles are in solid powder form. I want to study that effect of particles size on anticancer activity.

As you have prepared nanoparticles of bulk drug you need to either adsorb drug NPs on metallic or polymeric support or encapasulate drug inside nanocarrier so that you will get controlled release of drug and also dose reduction as compared to bulk drug alone.
Main advantage of nanopartilcle is dose reduction by controlled release and
avoid degradation of drug inside body
Raj, the concept of "nanoing" is much more than just improving solubility of poorly water soluble drugs.
When you convert ANY drug to its respective nanoparticles, due to enhanced surface area to volume ratioof nano, the encapulsated drugs may behave very differently. The inner atoms (of drugs) are exposed, and so, the properties of the nanodrugs may greatly vary from that of bulk. But, in that case, its very difficult to predict the drug activity and comparing with its bulk counterpart. So, to reply your query in specific , let's assume, the properties of ur drug is not altered when it becomes "nano".
1. According to ur statement, the drug u r using is "poor" water soluble. How much poor it is? What's the solubility limit in the culture media?
2. If you are using concentration that is beyond the solubility limit of the drug, only then you will see a difference between nano and free drug. If you are loading very less concentration of drug in nanoparticle, and if that concentration is well soluble in the given volume of media, then you may not get any improvement in the antitumor efficacy, because both will have similar solubility limit.
3. In this case, if your drug is adsorbed on to the nanoparticle surface, is there is any chemical interaction between your drug and the nanocarrier? If the interaction is very strong (say a covalent conjugation), ur drug may not be released in the culture media. in that case, after the cellular uptake, the cellular enzymes need to break the interacting bond, only then ur drug will give some effect. only then u can compare with the bulk
4. If ur drug is encapsulated in the nanocarrier, are u sure all ur drug is released from the carrier? only then u can compare with the corresponding molar concentration with the free drug counter part.
