I have no experience in caco-2 cell culture, but I am trying to differentiate them (which should take 21 days) and every time I try to do it (3x), always by the 10th day I can see with naked eye some white dots on the bottom of the wells (I don't know if it is just cell aggregates) and then the dots seem to detach and form "styrofoam-like" particles floating on the medium. I dont know if it fungus contamination (the medium is clean, no turbidity) or a normal behaviour of differentiation?
Hi Edna, I have no experience in culturing Caco-2 cells, but what you describe sound strongly like a contamination of your culture with yeast. If available use an objective with at least 20x magnification and have a close look at your "styrofoam-like" prticles. Do they then look like round (more or less) struktures in a chain like order? Then it is yeast.
You can eliminate it with fungin, if the culture is very important.
