I have extracted Glucan Particles from Sacch.cerev. and tried to solubilize them in NaOH 0.05M, autoclaving the suspension. After that, the suspension becames brown and when I centrifuge to collect the supernatant (where soluble glucan should be) to precipitate the glucan and centrifuge again, the pellet is brown. Am I precipitating something else or this colour is normal? The impurities should be eliminated during extraction procedure which is also made with NaOH at high temperatures until the supernatant is clean.
