I have bought this primary cell line from ABM Goods. After culturing in T-25 ECM coated flask I have observed many dead and floating cells in the culturing vessel. Is this normal? Does anybody have similar experiences?
Do you see this cell death when cells are less confluent than in the image? If they are normal oral cells, when epithelial cells become confluent it is normal to see cell death.
Thank you very much for your answer. When they are less confluent the amount of dead cells are also less. I am attaching a picture of less confluent cells herewith. Can you tell me why specially the epithelial cells start to die when they are confluent?
Sure! As you are probably aware, cell death is a normal part of the epithelial cell cycle. Anytime something goes wrong, cell cycle check points kick in and the cell will fix itself or apoptose (which is why you see a small amount of cell death in the less confluent cells). When the cells reach confluency, they are still able to grow, but have less space to do it. Most likely you're seeing the result of the competition for space.
I'm assuming that these cells are immortalized, even though they are normal? Although I would still expect to see the same thing with primary oral cells. The turnover rate of epithelial cells in the oral cavity is fairly high, which could also account for the cell death.
Thank you for your comment. This are actually primary cells but bought from a company. Even this high apoptosis rate in higher confluence level is normal for this cell type then still it is bad news for me. After reaching the confluence they all die in couple of days! It is necessary for my experiment to have live cells with a confluent layer even after 7 days from the day of reaching confluence.
