11 December 2022 7 5K Report

Hello everyone,

I am preparing protein samples for western blot, it is to check for K+ ion channels expression (I already performed patch clamp recordings so I know the proteins are expressed in the HEK 293 cells)

I transfected HEK 293 cells following lipofectamine 2000 protocol, and prepared cell lysate to get the proteins, but the GFP only transfected cells were transparent greenish white color, is it normal? or does that mean my protein samples are degraded?

(all cells are co-transfected with GFP and the ion channel , but only the GfP only cells are like this)

This my 2nd time to repeat this experiment and last time I didn't get anything in the western blot (although the BCA protein assay was positive), so I am not sure what might be the reason.

I am quite new in this area so I appreciate any help or advice :)

Thank you
