For spectrophotometric analysis, could I use the standard curve for a specific solution every time, once it is constructrd? Is it necessary to construct the curve every time before starting of the analysis each time?
No the linearity of the curve should have been proven during the validation study (since y = mx + b).
I agree with Bruce Neagle, In addition to that you could use the linearity of the curve, If
- Maintained all other parameter as such in validated method.
- Sample analysis is simple
- free from complex extraction procedure
- Its in aqueous/organic media not biological matrix
- It is UV method then OK but sensitive spectrometric method (MASS spec.) then you can not use same linearity curve
- the sample concentration is within the validated linearity range
Yes you need to do it. Standard curve will help you to ensure the accuracy of reading.
