Dear Luc Harvengt it has been a recurrent questions that other colleagues have asked me regarding walnut micropropagation. As probably you know it's a species hard to be in vitro cultured. Among all phases, in vitro establishment is the most difficult stage. That's why, once a genotype respond to in vitro conditions, the purpose is to use these materials as much as we can, at least it was what I did. Thus, I've mantained since 2008 untill last year (I moved the last April to other job) several clones activelly growing in vitro, without any sign of declination, producing average multiplication rates above 5, reaching 2 genotypes mutliplicaion rates close to 9.
My question has been prompted by several examples where partners have included in their requests for service of plants propagation by tissue culture, the high requirements that are the rule for small-fruited species (typically strawberry), which seems particularly excessive for tree species that are very difficult to disinfect. And that also seems very excessive when the tissue culture protocol doesn’t include a callus phase callus, and/or don't use either high growth factor concentration nor particularly mutagenic ones. Would some one have any further comment?
