I am using the Inoue buffer for the preparation of competent cells. Can it be used after storage on 4 degrees? The references recommend the usage of freezed aliquots or fresh buffer every time.
Emile Alghoul Hi according to this protocol, Inoue buffer is a mixture of MnCl2, CaCl2 and KCl2 in PIPES buffer, pH 6.7.

As with all buffer solutions used in molecular biology it is important that they remain free of environmental bacteria. You could autoclave this buffer in ~100 ml aliquots and store at RT or keep it stored in a sterile tissue culture hood at RT.
You could also sterile filter or autoclave a 10X stock solution of this buffer, freeze and restore to 1X with sterile water.
