Is G418 inhibited by Pen/Strep?

I am transfecting cells with a vector that has NeoR gene, and I want to select them with G418.

The growth media contains Pen/Strep.

Gibco's datasheet (and few more places that seem to cite the same text) claims that Pen/Strep is a selective inhibitor of G418 and they should not be used together.

But I couldn't find another source for that.

Hi Anna,

I also often obtain resistant cell lines by G418 selection to create stable expression strains. In my case, I use Gibco's G418 reagent with Pen/strep, and the cells are killed without any problem.

I recommend that you consider the optimal G418 selection concentration for your culture method beforehand.
