You are right. If you have a descent expression of your marker you can clearly see it with the naked eye in the cell pellet. GFP expressing cells looks green-yellow like. Am working with several different fluorescent reporters (Cerulean, GFP, zsGreen, mCherry etc) and they are all visible in the pellet of cells, with good expression of the reporter.
Yes. If you have high expression, you will notice the green color in your flask or cell pellet, especially if you hold it up to the light. I have seen it before, too.
