Hi...I want to do immunofluorescence staining for junctional proteins in bEnd.3 cells. Can anyone suggest me if I need to coat the cell culture plasticware with collagen? I am seeding cells on glass cover-slips in a 24 well plate. Literature suggests coating with type I collagen on transwell semipermeable supports for permeability assays but whether coating is required for immunofluorescence is not clear. I know collagen coating is required for other brain endothelial cells. But I am confused if I should coat every plasticware with collagen or not for bEnd.3 cells.
