I'm working with iPSC-derived neurons right now and differentiating my neurons using the StemCell Forebrain Differentiation and Maturation kits. I induce my NPCs using StemCell Smadi kits and it works great for the 6 iPSC cell lines I've tested. My NPCs express SOX1, PAX6, and Nestin and exhibit the correct morphology.
For one cell line (the first one I tested) the differentiation and maturation kit worked great, even at really low densities, obtaining nearly 100% yield of neurons. Nevertheless, for the other 5 cell lines I obtain a mixed culture with a variable (high though) proportion of astrocytes. In addition, the neuronal precursors exhibited an astrocyte-like morphology when I started the neuronal differentiation.
At first, I thought that it may be a problem with my cell lines, but after trying the medium with cell lines that have been generated at the same time and lab that the one that successfully differentiated to neuron I think that maybe the problem is the medium.
Is anyone working with these kits? If so, can you share your own experience?
