Some of my stock plants leaves are not expanded well. I tried with different auxins. but it does not work. Does anyone have a different thought about new hormones?
If you mentioned the name of your plant, it is better to comment on it, but usually, the poor leaf expansion may be due to the density of explants in a container. Adding adenine sulfate to the culture medium is also effective.
Density of Explants in the Culture could be one reason as the Plantlets will not have adequate space for the leaves to expand .
Secondly, you may decrease the concentrations of Cytokinin because higher concentrations of Cytokinin might result thicker leaves with short interludes. Regards.
Dinum Herath for 'improve leaf expansion', did you mean to make them 'grow bigger' in the medium? My opinion is that small leaves are generally ok. Once move into soil, they will start growing better. Or, you want a 'big leaves' in tissue culture stage for a special purpose?
I have noticed that subculturing the shoots in medium having IAA or GA3 help in leaf broadening. Sometimes Kn individually and in combination with auxins or Adenine sulphate also help in leaf lamina broadening there are alsp reports regarding same phenomenon.
Sometimes transferring the cultures in liquid medium on shaker also helps.
However it all depends on plant species, but you can try it out.
You may try with Kinetin...I found it good in expansion for some woody species...BAP is less effective in leaf expansion but callus production from wounded side...but my advise for you is that, expanded leaves generally gave larger leaf area but they became relatively fragile like a thin layer of caramel...you must be extra careful when handling it especially during transfer process....
I feel that those expanded leaves are thinner (cross section)...not sure whether it is fleshy enough for your subsequent experiment or not
Although it depends on species, liquid media enhanced leaf expansion. I have tested the comparison between solid and liquid media on different plant species, most of the case leaves become bigger than solid media.
I haven't worked on hormones for a long time. But I was able to improve apple proliferation by adding chitosan. You can read my article on this. Maybe it will help you.

Leaf size and shape is correlated with climatic factors. Light intensity, aeration and nutrient availability, can influence leaf size and shape. You may try different amount and sources of nitrogen too.
