Depend on the size of your sample and what you look for. Based on my experience,  rotary microtome with a freezing stage works very well with sample smaller than 2-3mm in diameter, and this should be good for most microscopy work in ascomycetes.

You should not have any problem using the rotary microtome with a freezing stage. But what are you interesting on finding? Ascus development ?

Hi, Yes ascus development and just generally the structure of the ascomata, thickness of walls etc.  I was planning to use the freezing stage for quick and easy sections, but then paraffin or resin for more detailed studies.  Have you found paraffin or resin to be better than freezing?

Resin and paraffin are good, but I prefer resin LRW. You will get more section and more information in details. Also you could do Transversal and longitudinal section easier.

Paraffin due to thick section the detailed information is good but limited to few section.

I've had good results using paraffin, but it depends on how many ascomata you have and their size. If you don't mind the slightly thicker sections, paraffin is a pretty quick way to get great anatomical detail for most fungal fruiting bodies.
