I want to edit a SNP in a non-coding sequence. I can't use the CRISPR HDR approach or prime editing because the PAM or the binding site of gRNA in the ssODN or pegRNA would be changed. I need to edit the SNP, and only the SNP. Base editing is a good approach. In theory, there is a good guide RNA, but I found it by placing the negative strand of DNA in a 3'-5' orientation in PnB, Be-Hive, and BE-Designer (with eA3A-BE3 as base editor). Please, can you tell me if this gRNA is usable? Thank you!
