Hello, I was planning to modify 4 kb GFP plasmid for transient expression in plants.
I was going to add 4 kb gene and make 8 kb plasmid.
Our lab use plant protoplasts for experiments and when I transfect them with GFP plasmid, it's delivered and it works fairly well. Btw, the facilitater is PEG.
But when I proposed this idea to someone else, he really disagreed with my plan.
I am awfully curious whether deliverying 8 kb plasmid into protoplasts is that hard to make it.
Is 8 kb thattt big for a cell to uptake?????????????????????????????????????????????????????

Dear Hyunbae,
You didn't tell why that person was disagree. However, I have no experience for on working with plant cells, but using 8 kb plasmids in animal cells is possible and common.
