During Fluo-4 calcium influx assay in human fibroblasts, I find that adding 2mM Ca in Ca-free well doesn't increase fluorescence intensity (FI) even over 10min period, which is expected. However, 20mM Ca rapidly increases FI for 3mins before reaching a plateau (see attached graph).
So I want to know if 20mM extracellular Ca is toxic to cells or not? Should I be expecting a calcium influx with 20mM concentration? If so, then what is the mechanism of Ca influx induced by 20mM Ca?
