My lab is engaged in a project on creatin a candidate DNA-vaccine. As a spin-off of the main work we want to try intranasal immunization of mice with our experimental DNA-vaccines. However, no one in the lab ever worked with this way of the DNA-administration to animals.
There's a lot of information on the topic in the public papers, however I would like to hear from a person that has a personal expertize in intaranasal airborn DNA-delivery.
My questions:
- What is the best-working transfection reagent, protocol or chemistry (in your personal experience)?
- Will PEI or lipofectamine work well or there is a definite need to swith to a different transfection reagents, to mix with plasmid DNA before applying to mice i.n.?
