I want to run my E. coli isolates on a PFGE gel but am having a little trouble with the lysis step due to the capsule. Anyone have any advice on how best to remove this? Currently using in well lysis with a lysis buffer consisting of 100 μg/mL proteinase K, 50 mM Tris–HCl at pH 8, 50 mM EDTA and 1% N-lauroyl sarcosine with incubation for 2 hours at 55°C.

I'd also like to find a way to remove the capsule for other analysis such as resistance profiling with and without the capsule.
