I would like to measure levels of testosterone in mouse serum and testicular tissue. The mouse serum measurement is relatively straightforward and readily available. However, I am not quite sure about how to go about measuring levels of testosterone in testicular tissue. I do not have access to fresh tissue currently, but I do have frozen mouse testes that are ready to use.
What protocol/kit would you recommend I use for this?

Our previous labs have always estimated Steroids from Frozen serum including testosterone. So treat them as fresh tissue, follow the same procedure as you plan for fresh tissues. I can not suggest the method depend on you budget. Earlier time people used to use radioimmunoassays. Now more simple methods would be available
As Dr. Subhash C. Juneja said in the last that simple methods are available. Let me tell you that we have developed ELISA kit (under the leadership of Prof. T. G. Shrivastava, Ex Professor, Department of Reproductive Biomedicine, NIHFW, New Delhi) that is very sensitive, accurate and reliable for the measurement of testosterone level in the blood serum as well as tissue homogenates. You can use the ELISA kit of Elabscience. Protocol of this kit are very easy to use. I think tissue homogenate used in this method should b prepared in PBS.
