I made 400 L of MEM 199 medium (sigma powder cat M0393 Medium 199 ) and added extra 8 L of water. Some people saying that i over diluted this media and it wont be good. Please give your opinion. thanks
True, the concentration is 0.98 of the intended standard concentration. Whether you want to use it as you mixed, take 1.02 times the volumes prescribed to make up for the dilution, or remix will depend largely on the nature of the research and tolerances to concentrations and volumes. If the research affects lives and very close tolerances to obtain results, then best to remix. If you have advisor or the "some people" mentioned normally provide excellent advice, it is best to stick with them unless you are the ultimate expert. The problem comes when you get mixed signals. You will have to defend your actions, so make sure you have good reasoning and documentation. If you situation normally involves close scrutiny and perfection, I would just remix to the standard. If you need added replicates, perhaps run it both with the standard and also with the slightly diluted concentration to see if there are differences.
What's cheaper, tossing out the 400 L of MEM and starting over or trouble-shooting any potential issues from using it? Or, you add more of the powdered MEM medium 199 so the concentration is correct (assuming it can be filtered/sterilized/whatever a second time).
