I am planning to make a single time stock solution of β-NADPH (10mM, 100x concentration) from 100 mg desiccated powder and store its single-use aliquot vials at -20C refrigerator. I am a bit confused about buffer choice and its alkaline pH for dissolving β-NADPH.
I have to test recombinant aldoketoreductase at different pH values for optimization studies.
β-NADPH is a co-factor in this enzyme assay.
So, if I dissolve β-NADPH in Na-bicarbonate buffer as per SIGMA recommendation, will it affect the final enzymatic reaction pH? Is there any alternative solvent/buffer recommendable in this case? Thanks
