I would like to understand how to correctly determine the passage number of the cell cultures we are growing.
Our lab currently purchases cryo-preserved cells that were frozen at the end of the primary culture, and I would like to know if I would be plating P0 or P1 when I defrost them and plate them in flasks.

Hi Mouna.
I guess you are not aware about the passage number of you cells before they froze down?
I would write "P1" on the dish/plate after thawing them.
When I want to freeze down some cells, I am used to write on the cryovial the same passage I have on the disk before freezing them, and once I revival the vial from nitrogen, I add "one" to number of the passage number written on the cryovial.
