Hello, I want to ask please about a soil sample preparation for HPLC. I have tried digestion, but running samples using HPLC did not give me some good chromatograms. I believe that I need to use another method. Any idea please? I am mainly looking for vanadium in soil samples.

Are you crazy? Pure Vanadium is an element and has no UV/Vis spectrum. Did you try ICP? There is a phosphomolybdate colorimetric procedure for Phosphorus that can be adapted for Vanadium.

And who is talking about vanadium element? Once within environmental samples (Sea water and soil samples in my case) vanadium can never exist as an element, it is either in one of its many oligomeric forms or as a complex in case there is a chealating reagent in the medium.

Dear Iman: I put "Vanadium HPLC in google" and several interesting papers easy to access appeared, so maybe you can take a look at some of them. I could help you if you were using ICP-MS, but then you can only determine Total V, not any speciation, obviously.
