I estimated the protein concentration using Lowry method, to be 321 mg/ml. I need to add varying FPU/gm of these celullase for the enzymatic hydrolysis of lignocellulosic substrates.Please suggest me the above calculation for the conversion of FPU/ml to FPU/gm.
If you found FPU of the enzyme solution per mL then you do not need to have it per gram. You could add a desired amount of mL of the solution of the enzyme to the substrate (lignocellulose) to get a desired FPU per gram of lignocellulose.
If you really need to know it per gram of enzyme solution: weight one mL of the solution and then you will know the density. Then you could calculate FPU/g of the enzyme solution.

I understand that you have calculated FPU/mL and mg/mL of total protein content (Lowry's assay). Now to calculate the specific activity (FPU/mg), use this: (FPU/mL) / (mg/mL); you will get FPU/mg of the protein.
With the density you can know the FPU/gr … I remember that the density of the Cellic CTec2 is 1.15 g/mL.
By the way, the 92 FPU/mL for CTec2 is low … normally the cellulase activity for those enzymes is more than 150 FPU/mL.
If FPU is greater than 150 , there is indicate hydrolysis unit is greater than 1000U/g, is it necessary to do calculate FPU?
