LED treated plants gave lower concentrations on APX, SOD and CAT antioxidant activities than fluorescent light...what does it tell from the concentration of the enzymes? which is better?
the most importnat parameters is balance between ROS production and scavenger. Enzyme activity is only ROS scavenger in certain compartments (3 SOD, 3 different APX). The best conclusion you will made if you can measure balance of the ROS (H2O2 and superoxide accumulations) in different compartments. This parameter give you real level of oxidative stress.
Dear Yurii, there are several methods for determination of ROS "imbalance" in tissue. There are many kits what can measure ROS level. In addition, one can use also DCHBS/AAP method for H2O2 balance measure and XTT for superoxide.
Level of antioxidants enzyme activity level cant give precise information about stress level.
And, of course, type of lamp, light intensity, spectrum must be consider as well.
I know very well the chemistry behind of such tests. It would be really interesting to discuss what these tests do measure, but I don't think that this is a right place for such discussion.
My answer to the main question: "
What can I conclude from the experiment?"
First, you must well understand what your antioxidant tests measure before interpreting your results and making any conclusion.
I think the ratio of antioxidants to oxidants shows the cells situation.Measuring one of them can not help us to understand oxidative stress in cells clearly.Higher amount of antioxidants due to oxidants shows healthier cells.
@Arda Acemi , can you share me methods on antioxidant analysis for APX, SOD and CAT? I have the original papers but they are too complicated to be understood
