The concentrated methanolic extrat schould be first shaked with n-butanol. Then the n-butanol phase evaporated.The obtained material dissolved in small amount of methanol and precipitated with ether.Ether should be 5 times of the concentrated methanolic extrat. You can use Sephadex LH-20 or Diaion for purification.You have to know that the methanolic extract contains not only saponins but other glycosides as flavonoids.

please Dr. ,I have already ppt saponin from butanol extract by acetone .then I dissolve the ppt in 80% aqueous methanol to solubilise it.I don't know which is better to isolate each saponin on sephadex or silica and what is the mobile phase

after precipitation , i filtered saponins using filter paper ..after a while the reidue turned yellowish brown and is really stuck to the filter paper,,,,any help?
