The elution buffer is

100 mM Tris(pH=8.0)

150 mM NaCl

1mM EDTA (This is not a metalloprotein)

I have adjusted the pH to 8.0.

I also have loaded 2 microlitre beads which clearly shows my protein of interest (confirmed with western blotting).

Please suggest.
