I get tumor tissue from the patients after the surgery. How to isolate the top layer of cells from a tumor?

I work on gastric cancer. I want to separate the top layer of tumor cells from the lower layers.

Though it's a little hard to differentiate grossly, the use of dye and observing surrounding tissue under the microscope will help to differentiate somehow.
