I detected two unequal concentrations of substrate with gradient concentration of inhibitor - can I fit a curve to obtain the Ki?

I have determined Km and Kcat of enzyme and want to determine Ki of an inhibitor,I just detected 2 different concentration of substrate with gradient concentration of inhibitor,can I use those data and fit curve to obtain the Ki?If I can ,which software to use?thx~

Dear Jia

Firstly, you have to identify which one is independent and which one is dependent variable. Then you can easily draw the line of concentration curve using Microsoft Excel or any statistical software like SPSS, SAS, R etc.
