Beta- mercaptaethanol is reducing agent thus it degrade the RNases, So i think you better add this after homogenisation also this may be helpful in retrieving RNA with good RIN value
I could hardly find the manufacture's protocol. Please attach the manual so that readers can assist you to suggest. Well, I use Qiagen instead, and the manufacture suggests users to add certain ratio of β-ME to their buffer prior to leukocyte lysing.
It is important to have basic knowledge of β-ME for RNA extraction. According to Qiagen, the agent "will irreversibly denature RNases by reducing disulfide bonds and destroying the native conformation required for enzyme functionality." But recent study conducted by Mommaerts et al. (2015) suggested replacement of β-ME with less toxic agent.

Yes, it needs to add mercaptoethanol and PLANTaid before mixing the homogenate. I have seen this "Biozol plant total RNA extraction kit" from Bioflux (Bioer).. Sorry to answer an old thread
