I am trying to solve for the amount of acetaminophen in one tablet of Tylenol. The experiment was done with a UV-VIS spectrometer where 4 standard solutions were prepared (4,6,8,10 ppm). The standards were plotted Absorbance vs. Concentration and a calibration curve was constructed with a best-fit line equation of y = 0.058x + 0.0452. After the standard absorbances were found and the best fit line equation was obtained the sample of acetaminophen was ran through the instrument and the absorbance was found. The absorbance was then plugged into the best-fit line equation and a concentration of 9.88 ppm was calculated for the sample. I am trying to find the amount of acetaminophen in the tablet and compare it to the label which states that there is 325 mg of acetaminophen in each tablet. Please help.
For this, first of all you have to decide in which release media (saliva, gastric, intestinal fluid, etc.) you want to find its amount. Once decided, you just put fixed number of tablets in that release media either directly or in membrane depending on the solubility of your drug and go on taking its absorbance with time using UV-Visible spectroscopic technique until you get the constant readings. Meanwhile you have to take the calibration plot of acetaminophen to get the value of molar absorptivity. Finally, by using Beer-Lambert's Law, you can easily find the amount of acetaminophen in your tablet. You can refer to this publication doi.org/10.1016/j.foodchem.2021.131925
