i need to know how to do a mic whit no soluble agent, I looked about quartenarium amonium MIC
but my big problem is that my solut is a gel and i cant to control contact surface if i made a coating in the well.

In my opinion try to dissolve your sample in DMSO and then subsequent dilution can perform in water. may be this information will helpful for you.

thanks, we have 3 agents which is impossible to disolve in DMSO. we are going to use dicloromethane.
If you want to perform your test in aqueous medium, you can try to start with a much lower concentration of you compound and see whether it is soluble at that particular concentration. Since your compound is a quaternary ammonium compound, it is supposed to be quite active as antimicrobial agent. So you can start with a much lower concentration for the determination of your MIC.
