Hi,

In my experiments I'm trying to induce apoptosis in cancer cells stably expressing HSV-TK by treating them with GCV.

I observe a significant decrease in treated cells compared to control cells 5 days after adding GCV. However, the treated cell number never drops under the originally seeded amount of cells.

In my understanding of how HSV-TK works all those cells should eventually undergo apoptosis, as they are all EdU positive, and I was wondering if anyone has an idea why they stay alive?

And does anyone have an idea why it takes up to 5 days after exposure for a difference to be visible? These cells are still happily proliferating after being treated with GCV which shouldn't be possible?
