Hi, I can differentiate primary white preadipocytes to mature adipocytes in vitro. At day 8 of differentiation I want to separate the cells that differentiated (lipid inclusion) from the undifferentiated cells. I usually trypsinize the cells and after centrifugation most cells can be pelleted. When I resuspend the pellet carefully in PBS an spin at 300g for 10 minutes I can see a floating layer assuming these are intact adipocytes. I think this is not the best method. Do you have any suggestion for a better method? Percoll?
