I'm doing experiment with N-acetylgalactosaminyltransferase 3 (GalNacT3) on endometrial cancer ( antigen retrieve with citrat buffer pH 6, 120 degree, 3 minutes; blocking H2O2 in 5 minutes; fist antibody 30 minutes ). The result showed so strong unspecific background staining. Does anybody have idea about how to reduce it? Should I change to another method of antigen retrieval? ( cause I've read about low pH buffer can reduce the strong background staining
