My r-protein (in e.coli) is only solublized in urea and I want touse it in a downsteam immunization procedure, dentaured protein is not convenient for production of Ab, how to solve this problem ?
Need to run a folding procedure first, you need to get rid of urea as slow as possible. You can use dialysis, dilutions, on-column folding, etc. No matter the selected method folding is tricky and depends on several variables such as your protein and the solvent you are using for folding.
After having done the refolding step (and getting rid of urea) as described above by @OmarOrtega, you need to think about what adjuvant(s) and/or immunopotentiator(s) to include in your immunization in order to optimize the immune response you want. HTH.
