How to prepare a stock solution of Microcystin LR?

First, make a 100 uM stock solution with 99.5189 ug MC-LR into 1mL water.

Then, Final volume/(100 uM stock / desired dilution)

For example, 0.2 mL / (100 uM / 10 uM) = 0.02 mL of stock needed

So for 0.2 mL of a 10 uM solution add 0.02 mL of 100 uM stock to 0.18 mL diluent and just repeat for other concentrations

Dear Menuja, if you want a serial dilution from 100 uM to 75 uM, then 50 uM and 10 uM serially in 0.2 ml medium so first prepare your 100 uM stock as per the manufacturer guidelines of your compound.

Now from 100 um to 75 uM;

V1 = C2V2/C1 so 75* 200 ul (or 0.2 ml)/100 = 150 ul so 200-150 = 50 ul (take 50 ul medium and add 150 ul from the 100 uM stock tube) it will be now as 75 uM in 0.2 ml.

I am using here 200 as total volume (0.2 ml) to convert in ul so you will directly get amount in exact ul and no need to convert it.

Note that ratio or factor from 100 to 75 dilution is 1.33, ratio from 75 to 50 dilution is 1.5 and ratio from 50 to 10 dilution is 5. So to check that above done calculation is right or not you can perform 2 simple steps to reassure as following;

200 (total volume) /150 = 1.33 so same ratio is achieved

1.33*75 (new dilution) = 100 (dilution of original stock)

Now from 75 uM to 50 uM;

V1 = C2V2/C1 so 50*200/75 = 133.3 ul so 200-133.3 = 66.7 ul (take 66.7 ul medium and add 133.3 ul from the 75 uM stock tube) it will now be as 50 uM in 0.2 ml.

Calculation Confirmation: 200/133.3 = 1.5 so same ratio is achieved

1.5*50 (new dilution) = 75 (stock dilution)

Now from 50 uM to 10 uM;

V1 = C2V2/C1 so 10*200/50 = 40 ul so 200-40 = 160 ul (take 160 ul medium and add 40 ul from the 50 uM stock tube) it will now be as 10 uM in 0.2 ml.

Calculation Confirmation: 200/40 = 5 so same ratio is achieved

5*10 (new dilution) = 50 (stock dilution)

It is one way you can easily perform serial dilutions from a certain stock.
