Dear colleagues!
I am in desperate need to do overlay data collected on different days in FACS Diva (need to compare some markers expression levels on different days of cell growth). The settings of data acquisition are the same, so overlay should be adequate. But the problem appears: Diva closes the "book" of opened experiment when you try to open another experiment in order to perform overlay. Is it possible to overcome this obstacle inside Diva or is there any third-party software where I can import fcs files from Diva and finally enjoy my overlay?

Hello! I also previously worked with FACS Diva software. However I do all the data export as .fcs files and then import them into Flowjo. There you can create your work sheet from whatever .fcs files you load into them (each vial has its own .fcs file). So you can easily overlay results gathered on different days. Flowjo takes a day or two to get used to and its best if someone can show you in person but in general the software is easy to handle.
Besides the raw data setting using software or readings obtained from different gated populations in the plots, one can also use ImageJ software to overlay flow cytometry data images if the user have kept the Gates identical throughout the set of experiments. Thanks, All the best.
