Soluble and immobilized enzyme, both are active at 500mM buffer concentration. But a higher concentration produces high noise during conductive measurements. On lowering the concentration to 50mM or upto 10mM, enzyme remain active. So which concentration can be considered to be optimum one for conductive measurements (50mM or 10mM).
If I well understand your question, you are doing conductometric measurements to monitor enzymatic activity. If that is correct, you should do optimisation. I mean use the two concentrations and measure for each your response and see which one will give you better sensitivity. I hope this would help.
You measure conductivity? then you should use the lowest buffer concentration (10 mM). I and my colleagues use 5 mM PBS buffer.The reason is that due to some complicated physical laws our conductometric devices give no response (signal) at buffer concentratios above 50 mM. Thus the lower buffer concentration gives higher biosensor signals.

I think you should proceed electrochemically. Square wave voltammetry, allowing you to determine the optimal concentration of the buffer, while calculating the limit of deviation.
