I want to lable mesoporous silica nanoparticles with the 5(6)-Carboxyfluorescein (FAM) dye.
Do you have any suggestions about the following:
- What is the functional group of FAM? Is it's carboxyl group?
- Is it better to add dye during MSNs synthesis or after synthesis?
- Is it necessary to add any functional group on the surface of MSNs?

Silica NPs have silanol (Si-OH) functional groups on their surface which can react with the carboxylic groups of 5(6)-carboxyfluorescein. In the suggested reference the silica NP precursor is first covalently attached to 5(6)-carboxyfluorescein by activation with dicyclohexylcarbodiimide (DCC).

-for nanoparticls of silica ,you can used reaction called (Sol-gel ).
-(SiO2 + KOH or K2CO3) + Fluorceine or its derivates.To this mexture added drop wise H2SO4 SOLUTION ( 98%).
finally washed product with distiled water.
