Mice colon has many different immune cells, I need to isolate the neutrophils, macrophages from colon, mucosa, does any one know the protocol regarding this method? Great thanks!
There are a few methodologies out there regarding leukocyte isolation from the gut. I've only worked with primate tissue however most protocols out there use some variation of EDTA media isolation followed by digestion with collagenase/other enzymes. My suggestion after experience with this is unless you're specifically interested in the epithelial layer vs the lamina propria is to combine your two isolated populations. To further isolate cells you will have to use discontinuous gradient centrifugation and possibly antibody selection for specific cell types. Luckily you're working with mice which have a lot of very good antibodies to work with.
A problem area you may encounter in the colon is the presence of excess mucus which may impair your isolation and selection. Let me know if you have other questions.
