I have this assignment and I'm struggling to interpret this graph. Could someone please help me understand why it also helps to understand the mammalian-1-hybrid assay?

I just need to know how each is expressing.
You need to give more context eg. what is the difference between ROS (reactive oxygen species?) and ROSI. Also, that graph lacks error bars indicating a lack of replicates.

So just to make sure I am interpreting your graph correctly. When you have just your vehicle and Gal-Gal you have no expression indicating that it is suppressing your promoter. When ROS is added to an empty vehicle there is no expression because there is no promoter or luciferase CDS to express. Your vehicle alone without the Gal-Gal has a leaky promoter causing weak luciferase expression while the vehicle with ROS causes strong expression indicating that it is an inducer for the promoter?

I think so yes. vehicle + gal empty us showing nothing, but I think you are right about the vehicle and rosi reaction being the inducer for the promoter. I'm not sure how to validate this answer, and not sure how to research for this.
Start by looking up your plasmids. Check what promoter they use. Look up the chemical name of your drug.
