I did automated DNA extraction from cervical swabs samples and for this is did elution in 100 microliter but at the end of DNA extraction I got very low DNA concentration which is like 15ng/microliter and I need 40 to 50 ng/microliter. Is there is any other method to increase the concentration of DNA in the samples? Does vacuum centrifuge work for this?
Ethanol precipitation will do and then you can resuspend your DNA pellet in a smaller volume.

I have not worked with this device.but if you can reduce the elution volume to 50ul, your concentration will increase to 2 times the current concentration.

Hello you have several aliquotes of extracted DNA you can combine them and centrifuge to collect DNA.
Or more better solution is DNA concentrator vacuum evaporator by which you can increase the concentration of DNA in your sample by losing volume l.

1- Do the elution step by using a lower than 100 micro may be used a 35 micro (if you can) in twice better than 100
2- Sometimes the critical point is an elution step ( the duration of waiting before centrifugation ) that depending on the kit type, but I think you need more than 3 min at least to get more percentage.
3- Fresh culture(overnight incubation) would be better than the old culture.

To increase the concentration you can use a low elution volume in the final step of extraction, for example 30 or 50ul
