To extract protein from plant tissue using chloroform/methanol method, at least 200 mg sample is required. However, root weight of few wheat seedlings (14-30 days) is not that much. Therefore, may I bulk the root sample of the same genotype at same replication to increase the sample weight? In such situation, is there any other way to improve the sample weight for the method?
You can increase the number of samples grown. Of course, applying dehydration stress to the specimens may increase the volume and weight of the root, but it can also cause experimental errors.
I think adding to nitrogen in planting will increase the protein

AOA... U need first to provide that nutrients which is a rich source of nitrogen because it's important things for protein accumlation... But one professor ans to u about dehydration I think it's not good because it ia dought stress and very dangerous... May be I am wrong but this my suggestion to u thanks....
