I need to expose HepG2 cells to AZT for 2 weeks to study its effects on telomerase expression. The literature suggests to subculture them weekly, but doesn't say what kind of surface they were grown on (flask or plates). Is it possible to grow these cells in 6-well plates for 7 days? The plated density is 5000 cells/cm^2.

Add/seed very little cells, in my opinion add 10-15 thousand cells/ well in 6 well plate to grow them for 7 days. Even you can try as little as 5 thousand, however, sometimes too little cells pose proliferation issues.
The surface preferably used are flasks and not plates. The cell density ideally used according to me shoud be around 8000 to 10000 cells per cm2.

I think you may seed cell number based on doubling time of HepG2 cell, than discard and fill up the complete medium as usual.
